Data dive on half lives and receptor binding kinetics

Hey everyone I have been spending my evenings digging through old literature trying to map out actual receptor binding affinities and real elimination half lives for some of our common research subjects. Most vendors list generic numbers but the actual lab studies show wild variance depending on the delivery method and local tissue concentration. I am particularly looking at subcutaneous versus intramuscular absorption rates in murine models. Has anyone here crunched the numbers on time to peak plasma concentration for the standard fragment sequences versus full chain variants? Drop your findings or spreadsheet notes if you have them.

Honestly most of these published papers are funded by groups with specific agendas. I would take half of those binding curves with a massive grain of salt.

I ran some basic calculations on molecular weight versus diffusion speed last month. Smaller chains definitely move faster through local tissue.

Check out the old pharmacokinetics textbooks from the nineties. They have great models for subcutaneous depot absorption.

If you want real data you have to run your own HPLC testing and stop trusting third party sheets blindly.

Total agreement with the skeptic above. Half the time the purity on the COA does not even match the mass spec results anyway.

Thanks for that tip, I will hunt down those texts. Do you know if they cover peptide degradation rates in interstitial fluid as well?

That matches my observation kineticmind. The smaller fragments seem to clear out of the local injection site way quicker.
